Pus10 is a pseudouridine synthase responsible for the formation of Ψ54 and Ψ55, but it may also modify additional targets: sR658-U21, a site in a structured small RNA with similarity to tRNA’s T-arm, is a potential non-tRNA substrate of Pus10. Previous in vitro studies showed that Cbf5 can catalyze Ψ55, but in S. islandicus, in vivo data indicate that Ψ54 and Ψ55 are synthesized exclusively by Pus10, with no compensatory role of Cbf5. Alongside Pus7 (Ψ13) and Cbf5 (H/ACA-guided modifications), Pus10 completes the set of core pseudouridine synthases in S. islandicus.
